Solutions for sample fractionation and semi-preparative GPC/SEC
GPC/SEC is not only used for molar mass determination, but also to fractionate samples:
- There are many applications which only require small-scale (few grams) sample fractionation. For example, for enrichment/cleaning of samples during organic synthesis or preparation of samples for supplementary analytical investigations, e.g. infrared spectroscopy, NMR, mass spectrometry or elementary analysis.
These requests can be effectively handled with a semi-preparative GPC/SEC system in a straightforward manner. The columns used for these applications are semi-preparative GPC/SEC columns filled with typical highly crosslinked material. The fractionation is performed under high pressure. - Clean-Up GPC systems are used for the fractionation of several (hundred) grams of material. This is a widely used technique for the purification and analysis of synthetic and biological polymers, such as nucleid acids, poylsaccharides or proteins. The columns used for these appications are usually filled with dextrane or
argarose material, the fractionation is performed under
low pressure.
PSS products and services for sample fractionation and semi-preparative GPC/SEC
PSS offers complete turn-key systems for semi-preparative GPC/SEC as well as GPC/SEC semi-preparative fractionation service analysis in our analytical service department. Please contact us for a detailed quote.
The turn-key system is based on the modular SECcurity GPC System including convenient automated fraction collection with the SECcurity fraction collector operated by WinGPC UniChrom GPC/SEC software. If the fractionation is automated, fraction marks are added to the WinGPC UniChrom raw data window and saved with the data for highest traceability.
Preparative GPC/SEC columns are the heart of the semi-preparative GPC/SEC system. PSS offers preparative columns with a wide range of column materials for aqueous and organic solvents alike. PSS preparative columns can be added to any LC system to transfer an analytical system into a semi-preparative system.
Theoretical background sample fractionation and semi-preparative GPC/SEC
What is the difference between a conventional GPC/SEC and a semi-preparative GPC/SEC?
In principle any analytical GPC/SEC system, that can handle higher flow rates and injection volumes, can be converted into a semi-preparative system. The analysis conditions for the two systems are compared in the following table.
|
Conditions |
analytical |
semi-preparative |
|
Column dimensions [mm]: |
8x300 8x600
|
20 x 300 |
|
Optimum flow rate: |
1 mL/min |
6 - 8 mL/min |
|
Injection volume: |
20-100µL |
up to 1mL |
|
Sample concentration: |
up to 3 - 5 mg/mL |
up to 10 mg/mL |
Usefull additional options for the semi-preparative system are the fraction collector and automated control using a powerful software package.
